چکیده

Soil microcosms are a valuable tool for the study of genetic interactions between microorganisms in natural soil environments. However, many factors, some of which are better studied than others, can affect gene transfer events occurring in soil systems, and hence should be carefully considered when performing experiments in soil microcosms. This paper is a guide to setting up and evaluating gene transfer experiments, and in particular conjugation studies in soil microcosms. Particular emphasis is given to the transfer of broad-host-range (primarily clinically derived) plasmids, and natural antibiotic resistance and catabolic soil plasmids. z 1998 Federation of European Microbiological Societies. Effect of tetracycline residues in pig manure slurry on tetracycline-resistant bacteria and resistance gene tet(M) in soil microcosms.

Effects of tetracycline residues from pig manure slurry on the prevalence of tetracycline-resistant bacteria and the tetracycline resistance gene, tet(M), were studied in soil microcosms. Four types of soil microcosms were established for a period of 152 days, supplemented with combinations of pig manure slurry and a tetracycline-resistant Enterococcus faecalis, CG110, containing the tetracycli...

Behavior of various hosts of the IncP-7 carbazole-degradative plasmid pCAR1 in artificial microcosms.

pCAR1 endows carbazole-degrading ability on its host. Three different host Pseudomonas (P. fluorescens Pf0-1, P. resinovorans CA10dm4, and P. chlororaphis IAM1511) were prepared to compare their behaviors in artificial microcosms (sterile soil and water supplemented with carbazole) with previous results for P. putida SM1443 (Shintani et al., Appl. Microbiol. Biotechnol., 80, 485-497 (2008)). We...

Mycelia as a focal point for horizontal gene transfer among soil bacteria

Horizontal gene transfer (HGT) is a main mechanism of bacterial evolution endowing bacteria with new genetic traits. The transfer of mobile genetic elements such as plasmids (conjugation) requires the close proximity of cells. HGT between genetically distinct bacteria largely depends on cell movement in water films, which are typically discontinuous in natural systems like soil. Using laborator...

Thematic Issue on Horizontal Gene Transfer Transformation of Acinetobacter baylyi in non-sterile soil using recombinant plant nuclear DNA

To provide estimates of horizontal gene transfer from transgenic crops to indigenous soil bacteria, transformation frequencies were obtained for naturally transformable Acinetobacter baylyi BD413 using a chromosomally integrated plant transgene. The transgene comprised sequences for two phenotypic markers: kanamycin resistance (npt II) and green fluorescent protein (gfp), expressed from their o...

Effect of Flooding and the nosZ Gene in Bradyrhizobia on Bradyrhizobial Community Structure in the Soil

We investigated the effects of the water status (flooded or non-flooded) and presence of the nosZ gene in bradyrhizobia on the bradyrhizobial community structure in a factorial experiment that examined three temperature levels (20°C, 25°C, and 30°C) and two soil types (andosol and gray lowland soil) using microcosm incubations. All microcosms were inoculated with Bradyrhizobium japonicum USDA6T...
